Mitotane (2,4'-DDD) is an antineoplastic agent used to research adrenocortical carcinoma. This isomer of DDD and derivative of dichlorodiphenyltrichloroethane (DDT) primarily exerts its adrenocorticolytic effect through lipotoxicity induced by intracellular free cholesterol accumulation. It also has direct pituitary effects on corticotroph cells and can induce CYP3A4 gene expression via steroid and xenobiotic receptor (SXR) activation.
