ML382 is a potent and selective positive allosteric modulator of MRGPRX1 (Mas-related G protein-coupled receptor X1, MrgX1), with an EC50 of 190 nM. It is intended for research use only and not sold to patients. In vitro, ML382 dose-dependently increases the potency of BAM8-22, a MRGPRX1 agonist. In vivo, ML382 increases inhibition of ICa by BAM8-22 in DRG neurons from MrgprX1 mice, enhances the inhibition of spinal synaptic transmission by BAM8-22, attenuates heat hypersensitivity, and inhibits nerve injury-induced ongoing pain in MrgprX1 mice.
