MOLTOX FT tests measure the ability of test material treatments to induce reversion of histidine-requiring Salmonella typhimurium strains or Escherichia coli tryptophan auxotrophs to their respective prototrophic conditions. The bacterial strains used in these assays are identical to those in conventional plate incorporation assays, described by Mortlemans & Zeiger, Mutat. Res. 455: 29, 2000 & Mortlemans & Riccio, Mutat. Res. 455: 61, 2000. The experimental design used in the MOLTOX FT tests is based on Gatehouses's adaption of the design reported by Luria & Delbruck, Mutat. Res. 53: 289, 1978 and Genetics, 28:491, 1943. The MOLTOX FT "471" Mutagenicity Assay kit contain the products required to perform a microtiter fluctuation test using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537 & an E. coli WP2 uvrA strain. This kit includes sufficient components for 1 test material in triplicate analysis of 6 concentrations, positive and negative controls, with & without S9.
