NecroIr2 is an iridium(III) complex that acts as a necroptosis inducer in Cisplatin-resistant lung cancer cells (A549R). It selectively accumulates in mitochondria, causing oxidative stress and a decrease in mitochondrial membrane potential (MMP). NecroIr2 activates receptor-interacting serine-threonine kinase 3 (RIPK3) and mixed lineage kinase domain-like pseudokinase (MLKL), and also regulates CDK4 expression.
- Induces necroptosis by activating RIPK3 and MLKL, and regulating CDK4 expression.
- Over 90% accumulates in mitochondria in A549R and CFSE labelled L02 cells.
- Leads to increased ROS generation and loss of mitochondrial membrane potential.
- Induces necroptosis by arresting the cell cycle at the G0/G1 phase in a dose-dependent manner.
- Inhibits the proliferation of A549R cells in a dose-dependent manner.
- Appears as a brown to reddish brown solid.
- Soluble in DMSO at 50 mg/mL.
