NSC 194308 is an enhancer of U2AF2-RNA complexes, increasing the association of the U2AF1-U2AF2-SF1-splice site RNA complex by binding between the RRM1 and RRM2 motifs of U2AF2. This compound inhibits pre-mRNA splicing by stopping spliceosome assembly when U2AF recruits U2 snRNP to the branchpoint, and also enhances pre-mRNA binding to U2AF2, leading to selective cell death in leukemia cell lines with spliceosome mutations.
