PACAP (1-38) free acid is a desamido-PACAP (1-38), which is human, ovine, and rat. It activates the human PAC1 receptor and induces cyclic AMP (cAMP) generation in both NS-1 neuroendocrine cells and non-neuroendocrine HEK293 cells. In vitro, it promotes neuroendocrine cell differentiation in NS-1 cells with an EC50 of 10.6 nM and induces cell growth arrest in NS-1 cells at concentrations of 0.1-300 nM.
