Pilaralisib is a potent and highly selective class I PI3Ks inhibitor with IC50s of 39 nM for PI3Kα, 383 nM for PI3Kβ, 23 nM for PI3Kγ, and 36 nM for PI3Kδ. It is also known as XL-147 and SAR245408.

• Potent and highly selective class I PI3Ks inhibitor.
• Inhibits PI3Kα, PI3δ, and PI3Kγ with IC50s of 39, 36, and 23 nM respectively.
• Less potent against PI3Kβ (IC50 of 383 nM).
• ATP-competitive inhibitor with a Ki value of 42 nM for PI3Kα.
• Weak inhibitory activity toward VPS34 (IC50 of approximately 7.0 μM) and DNA-PK (IC50 of 4.75 μM).
• Does not inhibit mTOR activity (IC50 > 15 μM).
• Inhibits EGF-induced PIP3 production in PC-3 and MCF7 cells with IC50s of 220 and 347 nM.
