Pralsetinib (BLU-667) is a potent, selective RET kinase inhibitor. It targets wild-type RET and various mutants (V804L, V804M, M918T) and the CCDC6-RET fusion, with IC50s from 0.3 to 0.4 nM. For research use only, it inhibits NSCLC and thyroid cancer xenograft growth driven by RET mutations and fusions, without inhibiting VEGFR-2. Dose-dependent activity is seen in KIF5B-RET Ba/F3 and V804L Ba/F3 allograft tumors.
