This gene encodes a member of the TNF cytokine family, acting as a ligand for osteoprotegerin and playing a key role in osteoclast differentiation and activation. It also functions as a dendritic cell survival factor and regulates T cell-dependent immune responses. T cell activation induces expression of this gene, promoting osteoclastogenesis and bone loss. The protein activates the antiapoptotic kinase AKT/PKB via a signaling complex with SRC kinase and TRAF6, suggesting a role in apoptosis regulation. Disruption of the related gene in mice causes severe osteopetrosis, lack of osteoclasts, and defects in T and B lymphocyte differentiation, as well as failure to form lobulo-alveolar mammary structures during pregnancy. Two alternatively spliced transcript variants have been identified.
