SZM679 is a potent, orally active, and selective RIPK1 inhibitor with Kd values of 8.6 nM for RIPK1 and >5000 nM for RIPK3. It reverses tumor necrosis factor-induced systemic inflammatory response and decreases Tau hyperphosphorylation, neuroinflammation, and RIPK1 phosphorylation in the hippocampus and cortex. SZM679 can be used in research for Alzheimer's disease (AD). For research use only.
- Exhibits anti-necrosis activity by inhibiting the RIPK1 pathway with an EC50 value of 2 nM.
- Protects against TNF-α, cycloheximide, and z-VAD-fmk (TCZ)-induced necroptosis.
- Selectively inhibits RIPK1 expression and blocks necrosome formation.
- Improves cognitive function and rescues brain structure damage in AD mice.
- Decreases AD biomarkers and inflammatory cytokine levels in brain tissues.
