Upacicalcet sodium is a non-peptide calcimimetic agent that functions as a calcium-sensing receptor (CaSR) agonist. It is used in secondary hyperparathyroidism (SHPT) research due to its ability to decrease serum intact parathyroid hormone (iPTH) and calcium (Ca2+) levels, which helps reduce hypocalcemia and gastrointestinal complications. This compound also improves vascular calcification and bone disorders in models of adenine-induced SHPT and inhibits cortical pore formation and bone fibrosis in rats with chronic kidney disease (CKD).
- Functions as a CaSR agonist (EC50 = 10.8 nM)
- Increases intracellular Ca2+ and IP-1 in human CaSR-expressing cells
- Reduces serum iPTH, Ca2+, and Pi levels in rats
- Inhibits parathyroid hyperplasia in CKD rat models
- Inhibits ectopic calcification and cortical hole formation in CKD rat models
