This gene belongs to the PDGF/VEGF growth factor family and encodes a heparin-binding homodimeric protein essential for angiogenesis. It promotes vascular endothelial cell proliferation and migration. Disruption in mice impairs embryonic blood vessel formation. The gene is upregulated in many tumors, correlating with tumor stage, and is elevated in POEMS syndrome. Variants are linked to diabetic microvascular complications and atherosclerosis. Multiple isoforms arise from alternative splicing, translation initiation, and stop codon readthrough; one extended isoform is antiangiogenic. Expression is also regulated by an upstream ORF within an IRES. VEGF levels rise during SARS-CoV-2 infection, promoting inflammation and a feedback loop involving Ang II and cytokine release.
