Cytokeratins comprise a diverse group of intermediate filament proteins (IFPs) that are expressed as pairs in both keratinized and non-keratinized epithelial tissue, where they constitute up to 85% of mature keratinocytes in the vertebrate epidermis. Cytokeratin 7 is expressed in epithelial cells of ovary, lung and breast. It is often used in conjunction with cytokeratin 20 and CDX-2 in distinguishing pulmonary, ovarian and breast carcinomas (CK7+) from most colon carcinomas (CK7-).

Mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC). Initially, inherited mutations in the MSH2 and MLH1 homologs of the bacterial DNA mismatch repair genes MutS and MutL were found at high frequency in HNPCC and were shown to be associated with microsatellite instability. The demonstration that 10 to 45% of pancreatic, gastric, breast, ovarian and small cell lung cancers also display microsatellite instability has been interpreted to suggest that DNA mismatch repair is not restricted to HNPCC tumors but is a common feature in tumor initiation or progression.

Recognizes a protein of 120-80kDa, identified as E-cadherin. Cadherins comprise a family of Ca2+-depedent adhesion molecules that function to mediate cell-cell binding critical to the maintenance of tissue structure and morphogenesis. The c;assical cadherins, E-, N- and P-cadherin, consist of larg extracellular domains characterized by a series of five homologous NH2 terminal repeats. The relatively short intercellular domains interact with a variety of cytoplasmic proteins, such as β-catenin, to regulate cadherin function. E-cadherin plays an important role in epithelial cell adhesion. A decreased expression of E-cadherin is associated with metastatic potential and poor prognosis in breast cancer, prostate and esophageal cancer. In combination with p120 Catenin, it is useful for the differentiation between ductal (E-cadherin +) and lobular (E-cadherin -) breast carcinomas. It may also help in diagnosis of mesothelioma.
