fastStart(TM) Universal SYBR(R) Green Master (Rox) is a ready-to-use hot start reaction mix for qPCR and RT-qPCR on all real-time PCR systems requiring normalization with ROX. SYBR(R) Green I is a DNA double-strand-specific dye. During each phase of DNA synthesis the SYBR(R) Green I dye which is included in the reaction mix binds to the amplified PCR products. The amplicon can be detected by its fluorescence. Hot start protocols have been shown to significantly improve the specificity sensitivity and yield of PCR. Heat-labile blocking groups on some of the amino acid residues of fastStart(TM) Taq DNA Polymerase make the modified enzyme inactive at room temperature (15 to 25C). Therefore there is no elongation during the period when primers can non-specifically bind. fastStart(TM) Taq DNA Polymerase is activated by removing the blocking groups at a high temperature (i.e. a pre-incubation step at 95C).

High Salt-T4 DNA Ligase is a salt-tolerant mutant of T4 DNA Ligase that catalyzes the formation of a phosphodiester bond between juxtaposed 5? phosphate and 3? hydroxyl termini in duplex DNA or RNA. It is designed to function at higher salt concentrations than wild type T4 DNA Ligase. The catalytic reaction requires ATP as a cofactor. High Salt-T4 DNA Ligase will join blunt end and cohesive end termini at salt concentrations as high as 300 mM without any loss in activity, and can also repair single-stranded nicks in duplex DNA, RNA or DNA/RNA hybrids. At the same time, T4 DNA ligase closes the gaps in these DNA substrates.
