This gene encodes a cell adhesion protein from the immunoglobulin superfamily involved in cell-to-cell and cell-matrix interactions during development and differentiation. It plays a role in nervous system development by regulating neurogenesis, neurite outgrowth, and cell migration. Additionally, it aids in the expansion of T lymphocytes, B lymphocytes, and NK cells, crucial for immune surveillance. The protein interacts with fibroblast growth factor receptors, N-cadherin, and other extracellular matrix components, triggering signaling pathways such as FYN-FAK, MAPK, and PI3K. A prominent isoform, CD56, is implicated in myeloproliferative disorders like acute myeloid leukemia, with its expression correlating with disease progression. Increased CD56 expression is linked to poor survival in AML, while lower CD56-expressing NK cells are associated with severe COVID-19. Alternative splicing produces multiple isoforms.
