The EGF receptor family comprises several related receptor tyrosine kinases that are frequently overexpressed in a variety of carcinomas. Members of this receptor family include EGFR (HER1), Neu (ErbB-2, HER2), ErbB-3 (HER3) and ErbB-4 (HER4), which form either homodimers or heterodimers upon ligand binding. The gene encoding ErbB-4 is expressed as a full-length protein, which produces a short membrane-anchored cytoplasmic domain fragment and a long ectodomain fragment. The short fragment is heavily tyrosine phosphorylated and possesses tyrosine kinase catalytic activity toward an exogenous substrate. Proteolytic cleavage of ErbB-4 is promoted by the binding of heregulin. PMEPA1 (prostate transmembrane protein, androgen induced 1 is a 287 amino acid single-pass membrane protein that contains WW-binding motifs and localizes to the cell membrane. Expressed at high levels in prostate, kidney and ovary, PMEPA1 interacts with NEDD4 and may play a role in regulating AR (androgen receptor) levels, specifically in prostate cells. Down regulation of PMEPA1 is observed in prostate tumors, suggesting that PMEPA1 may exhibit activity as a tumor suppressor. Calbindin 1, also known as calbindin D28K, is a member of a family of calcium binding proteins. This protein family also includes calmodulin, calbindin 2, S-100, and troponin C. Calbindin 1 is a 261-amino acid protein with 6 EF-hand domains, 4 of which are active calcium-binding domains. Expressed in brain, ovary, uterus, testis, pancreas, liver, kidney and intestine, calbindin 1 acts as a calcium-buffering agent and alters the activity of the plasma membrane ATPase. In neuronal cells, calbindin 1 modulates calcium channel activity, calcium transients and intrinsic neuronal firing activity. This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. Ki-67 antigen is a nuclear, non-histone protein that is present in all stages of the cell cycle except G0. This characteristic makes Ki-67 an excellent marker for proliferating cells and is commonly used as one of the prognostic factors in cancer studies. A correlation has been demonstrated between Ki-67 index and the histo-pathological grade of neoplasms. Assessment of Ki-67 expression in renal and ureter tumors shows a correlation between tumor proliferation and disease progression, thus making it possible to differentiate high-risk patients. This antibody recognizes a protein of ~150 kDa, identified as Desmoglein-1 (DSG1). Desmoglein-1 is a member of the desmosomal cadherin family. Desmosomes are intercellular adhering junctions that represent cell surface attachment sites for intermediate filament. Desmocollins and desmogleins are the main desmosomal transmembrane proteins. Desmogleins consist of Dsg1, Dsg2, Dsg3, and Dsg4 isoforms. Within the desmosome, the extracellular domain of desmoglein is essential for calcium dependent heterophilic binding to the desmocollins, whereas the intracellular domain is essential for binding to the desmosomal plaque protein, plakoglobin.
