Biotium Cytokeratin 20 (KRT20) (Colorectal Epithelial Marker)(KRT20/1991), CF488A conjugate, 0.1mg/mL
This MAb recognizes an intermediate filament protein of 46 kDa, identified as cytokeratin 20 (KRT20). KRT is abundantly expressed in goblet cells and enterocytes of the gastrointestinal tract. It is a useful marker of pancreatic and colorectal cancer. KRT20 is expressed under normal, hyperplastic and neoplastic conditions. It has been detected in adenocarcinomas of the colon, stomachand biliary tract. Breast carcinomas are generally non-reactive. Eukaryotic histones are basic and water-soluble nuclear proteins that form hetero-octameric nucleosome particles by wrapping 146 base pairs of DNA in a left-handed supealpha-helicalturn sequentially to form chromosomal fiber. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. Thyroperoxidase (TPO) is a 933 amino acid, type I transmembrane glycoprotein that plays a key role in thyroid gland function and autoimmunity. It is present as a dimer on the apical surface of thyroid follicular cells. TPO functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA. Malignant thyroid tumors exhibit an anomaly in TPO resulting in lower affinity for anti-TPO. This antibody may aid in the differentiation between benign and malignant thyroid tumors. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol. Recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.4). It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. VISTA / GI24 is a transmembrane protein expressed in bone, on embryonic stem cells (ESCs), and on tumor cell surfaces. On ESC s, Gi24 appears to positively interact with BMP-4, potentiating BMP signaling and the transition from an undifferentiated to a differentiated state. On tumor cells, Gi24 both promotes MT1-MMP expression and activity and serves as a substrate for MT1-MMP. This increases the potential for cell motility. Mature human Gi24 contains a 162aa extracellular region with one V-type Ig-like domain and a 96aa cytoplasmic domain. Human Gi24 undergoes proteolytic cleavage by MT1-MMP, generating a soluble 30 kDa extracellular fragment plus a 25-30 kDa membrane-bound fragment. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). It is also expressed on the surface of normal early lymphoid progenitor cells, immature B cells within adult bone marrow and germinal center B cells within lymphoid tissue.
