Recognizes a 180-185 kDa protein, identified as isoform of leukocyte common antigen (CD45RO) (4th Leucocyte Typing Workshop: Code No. N31). The epitope recognized by this antibody is sensitive to neuraminidase digestion. This antibody reacts with mature activated T-cells, most thymocytes, and a sub-population of resting T-cells within both CD4 and CD8 subsets. It shows no reactivity with normal B or natural killer cells, but reacts with granulocytes and monocytes. Reportedly, it is useful to identify T-cell lymphomas and leukemias. Recognizes a 33 kDa glycoprotein, identified as Nucleophosmin (NPM). It is predominantly localized in the nucleus of cells in most tissues. NPM is involved in ribosomal assembly and rRNA transport. It is an abundant protein that is highly phosphorylated by Cdc2 kinase during mitosis. This phosphoprotein moves between the nucleus and the cytoplasm. It is thought to be involved in several processes including regulation of the ARF/p53 pathway. A number of genes are fusion partners, in particular the anaplastic lymphoma kinase gene on chromosome 2. Mutations in exon 12 affecting the C-terminus of the protein are associated with an aberrant cytoplasmic location. Mutations in this gene are associated with acute myeloid leukemia. Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. This antibody recognizes an epitope encoded by exon v6 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. This antibody recognizes a protein of 25-26 kDa, identified as Bcl-2. This antibody shows no cross-reaction with Bcl-x or Bax proteins. Bcl-2 suppresses apoptosis in a variety of cell systems including factor-dependent lymphohematopoietic and neural cells. It regulates cell death by controlling the mitochondrial membrane permeability. Bcl-2 appears to function in a feedback loop system with caspases. This antibody reacts with the first EGF repeat in the extracellular domain of Ep-CAM. Calcineurin B homologous protein 2, also known as hepatocellular carcinoma-associated antigen 520, is a 196 amino acid protein that contains 4 EF-hand domains and plays a potential role in transmembrane Na/H exchange. By binding to and activating NHE-1, CHP2 increases the pH and works to protect cells from serum deprivation-induced death. Though not typically detected in normal tissues, CHP2 is highly expressed in malignantly transformed cells and is therefore considered to be a tumor-associated antigen. Ectopic expression of CHP2 promotes proliferation of HEK293 cells and knockdown of CHP2 mRNA in HepG2 cells inhibits cell proliferation. Alpha actinins belong to the spectrin gene superfamily which represents a diverse group of cytoskeletal proteins, including the alpha and beta spectrins and dystrophins. Recognizes a protein of about 31 kDa, which is identified as PD-L2 (same as PDCD1LG2). Engagement of CD28 by B7-1 (CD80) or B7-2 (CD86) in the presence of antigen promotes T cell proliferation, cytokine production, differentiation of effector T cells and the induction of Bcl-x, a promoter of T cell survival. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk's and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Tumor necrosis factor (TNF) is a pleiotropic cytokine whose function is mediated by two distinct cell surface receptors, designated TNF-R1 and TNF-R2, which are expressed on most cell types.
