The B cell-specific moloney murine leukemia virus integration site 1 (Bmi-1) is a transcriptional receptor of the polycomb gene family involved in several cellular processes including cell-cycle regulation, apoptosis, and maintenance of adult and neoplastic stem cells by providing self-renewal capacity. Further, Bmi-1 expression has been associated with malignant transformation, tumor progression, metastatic disease, and poor prognosis in human malignancies. The protein encoded by this gene is a homeobox-containing developmental regulator associated with liver development. The encoded protein binds to the alpha-fetoprotein (AFP) gene promoter and increases the expression of AFP. This gene is overexpressed in some lung cancers and is linked to poor patient survival, possibly due to its resistance to cisplatin. This gene is aberrantly methylated in pancreatic cancer, deleted in squamous cell lung carcinomas, and acts as a tumor suppressor in esophageal cancer. This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14. B7-H4 (B7-homolog 4), belongs to the B7 immunoglobulin superfamily of ligand-lymphocyte interacting proteins. Expressed primarily on the membrane of lymphoid cells, B7-H4 is an immuno-inhibitory protein that interacts with receptors on the surface of T lymphocytes, thus mediating cellular and humoral immune responses. Overexpression of the B7-H4 protein is associated with certain malignancies, including ovarian and breast cancer, as its interaction with T cells suppresses tumor-associated immunity. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol.
