Recognizes 183 kDa protein with DNA-binding characteristics, which is identified as a myeloid specific antigen. It reacts with myeloid precursor cells and granulocytes in bone marrow. Its antigen appears to be restricted to M2 and M3 acute myelogenous leukemia (AML) subtypes. Markers of myeloid cells are useful in the identification of different levels of cellular differentiation. This MAb reacts with early precursor and mature forms of human myeloid cells. It is useful in the identification of myelogenous leukemias, distinguishing granulocytic sarcomas from lymphoid malignancies and also in the study of differentiation and transformation of human myeloid cells. Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody recognizes a transmembrane glycoprotein of 95 kDa, identified as CD18 or integrin -2 (Workshop III). It complexes non-covalently with either L, M, or X integrin (CD11a, b, or c) to form the heterodimers, LFA-1, Mac-1, and p150,95, respectively. LFA-1 is the receptor for three members of the Ig supergene family of proteins, ICAM-1 (CD54), ICAM-2 (CD102), and Mac-1 and p150,95 bind to ICAM-1, fibrinogen, and iC3b. ICAM-3 (CD50). CD18/CD11 heterodimeric molecules are involved with cell/cell and cell/extracellular adhesion in immune and inflammatory responses. This MAb recognizes a protein of 56.5 kDa, identified as cytokeratin 10 (CK10). CK10 is expressed in all suprabasal layers of the epidermis. In the epidermis, expression of CK10 strictly parallels the extent of differentiation; it is absent in the basal layer, appears in the first suprabasal layers and increases in concentration towards the granular layer. However, CK10 is rarely detected in early stages of vulvar squamous carcinomas (tumors less than 2 cm, clinical stage I) regardless of the tumor grade. In larger and more advanced tumors (greater than 2 cm, clinical stages II and III), CK10 is detected very frequently. This Ab reacts with the rod domain of human cytokeratin 19 (CK19), a polypeptide of 40 kDa. CK19 is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. In Western blotting, this antibody detects an antigen of 125 kDa in human liver and 135 kDa in tumors of histiocytic origin. Comparative study of this MAb and a standard CD68 MAb showed that their antigens are different. Its antigen in all macrophage types studied is located on the plasma membrane and within cytoplasmic structures including lysosomes. This MAb shows a restricted reactivity to cells of the monocyte/macrophage system. It specifically reacts with blood monocytes and stains resident macrophages in a wide variety of human tissues. This MAb does not stain antigen-presenting cells, e.g., Langerhans cells.
