The x-ray repair cross-complementing (XRCC) proteins are responsible for efficiently repairing and maintaining genetic stability following DNA base damage. These genes share sequence similarity with the yeast DNA repair protein Rad51. XRCC1 is a protein that facilitates the DNA base excision repair pathway by interacting with DNA ligase III and DNA polymerase to repair DNA single-strand breaks. XRCC2 and XRCC3 are both involved in maintaining chromosome stability during cell division. XRCC2 is required for efficient repair of DNA double-strand breaks by homologous recombination between sister chromatids, and XRCC3 interacts directly with Rad51 to cooperate with Rad51 during recombinational repair. XRCC4 is an accessory factor of DNA Ligase IV that preferentially binds DNA with nicks or broken ends. This MAb reacts with MUC1. The dominant epitope of this MAb has not yet been determined. MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and over expressed abundantly in 90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four c-erbB receptors and localize with c-erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. This antibody recognizes a protein of 145 kDa, identified as CD11c. CD11c (ITGAX), a member of the leukointegrin family, shares the same beta subunit with other members of the leukocyte adhesion molecule family, which includes CD11a (LFA-1), CD11b (MAC-1) and CD11d (ITGAD), but has a unique alpha chain. CD11c has been shown to play a role in phagocytosis, cell migration, and cytokine production by monocytes/macrophages as well as induction of T-cell proliferation by Langerhans cells. CD11c is expressed prominently on the plasma membranes of monocytes, tissue macrophages, NK cells, and most dendritic cells (DCs). A lower level of expression is also observed on neutrophils as a result of its high level of expression on most DCs. An antibody to CD11c may aid in identification of lesions with histiocytic origin. Immunoglobulins are four-chain, Y-shaped, monomeric structures comprised of two identical heavy chains and two identical light chains held together through interchain disulfide bonds. The chains form two domains, the Fab (antigen binding) fragment and the Fc (constant) fragment. Immunoglobulin D (IgD) exists as a monomer with light chains.
