The intestine-specific transcription factors CDX1 and CDX2 are important for directing intestinal development, differentiation, proliferation and maintenance of the intestinal phenotype. CDX2 protein expression has been seen in GI carcinomas. Anti-CDX2 has been useful to establish GI origin of metastatic adenocarcinomas and carcinoids and is especially useful to distinguish metastatic colorectal adenocarcinoma from lung adenocarcinoma. However, mucinous carcinomas of the ovary also express CDX2 protein. B-cell lymphoma/leukemia 10 (BCL10) is a signaling protein that promotes apoptosis, pro-caspase-9 maturation, and activation of NF-kappa-B via NIK and IKK. BCL10 contains an N-terminal caspase recruitment domain (CARD), which is found in a number of apoptotic regulatory molecules. BCL10 is involved in the adaptive immune response, and may be an adapter protein between upstream TNFR1-TRADD-RIP complex and the downstream NIK-IKK-IKAP complex. A BCL10 translocation is recurrent in low-grade mucosa-associated lymphoid tissue (MALT lymphoma). This antibody recognizes a 105 kDa phosphoprotein, identified as retinoblastoma (Rb) gene product. Its epitope is localized between aa 703-772. It shows no cross reaction with p107 or p130. It specifically stains the nuclei of BT-20 cells and primary human foreskin fibroblast (HFF) cells. It does not stain the Rb-negative BT549 cells. It reacts with the hyperphosphorylated as well as the un (under) phosphorylated form of the Rb protein. Retinoblastoma gene product plays a key role in cell cycle control. It has been identified as a tumor suppressor gene whose loss of its function leads to tumor development. ETS1 proto-oncogene is an important transcription factor that plays a role in cell proliferation and differentiation. ETS1 is related to the growth of carcinoma cells by its regulation of the transcription of matrix metalloproteinases and urokinase-type plasminogen activator. The processes of tumor invasion and metastasis depend on the increased proteolytic activity of the invading tumor cells that may involve matrix metalloproteinases, cathepsins B and D and plasminogen activator in the metastatic cascade. ETS1 is preferentially expressed in lymphoid cells, where it is essential for the maintenance of the normal pool of resting T and B cells. Renalase is a 342 amino acid FAD-dependent amine oxidase that is highly expressed in kidney and is expressed at a lower level in heart, skeletal muscle and small intestine. Renalase is secreted in the blood by the kidney and it is thought to regulate cardiac function and systemic blood pressure. It is also suggested that Renalase functions as a hormone that metabolizes circulating catecholamines, which have an active role in the sympathetic and parasympathetic nervous systems. Individuals with chronic kidney disease and end-stage renal disease have markedly reduced levels of plasma Renalase than healthy individuals. Infusion of Renalase in animal models causes decrease in heart rate, cardiac contractility and blood pressure. This antibody recognizes an epitope encoded by exon v4 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. The expression of specific cell adhesion molecule CD44 splice variants has been reported to be associated with metastasis in certain human malignancies. CD45R, also designated CD45 and PTPRC, has been identified as a transmembrane glycoprotein, broadly expressed among hematopoietic cells. Multiple isoforms of CD45R are distributed throughout the immune system according to cell type. These isoforms arise because of alternative splicing of exons 4, 5, and 6. The corresponding protein domains are characterized by the binding of monoclonal antibodies specific for CD45RA (exon 4), CD45RB (exon 5), CD45RC (exon 6) and CD45RO (exons 4 to 6 spliced out). The variation in these isoforms is localized to the extracellular domain of CD45R, while the intracellular domain is conserved. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). GP2 (glycoprotein 2), also known as ZAP75, is a 537 amino acid secreted protein. It is an integral membrane protein that is secreted from intracellular zymogen granules and associates with the plasma membrane via glycosylphosphatidylinositol (GPI) linkage. GP2 is cleaved and then released into the pancreatic duct along with exocrine secretions. GP2 binds pathogens such as enterobacteria, thereby playing an important role in the innate immune response. The C-terminus of this protein is related to the C-terminus of the protein encoded by the neighboring gene, uromodulin (UMOD). GP2 is also expressed on the apical plasma membrane of specialized microfold (M) cells among enterocytes and serves as a transcytotic receptor for mucosal antigens. This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation. The matrix metalloproteinases (MMP) are a family of peptidase enzymes responsible for the degradation of extracellular matrix components, including collagen, gelatin, fibronectin, laminin and proteoglycan. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. This antibody recognizes a protein of 31 kDa, which is identified as MTAP (5'-deoxy-5'-methylthioadenosine phosphorylase). It catalyzes the reversible phosphorolysis of methylthioadenosine, which is important in polyamine metabolism and for the salvage of adenine and methionine. The gene encoding MTAP is linked to the tumor suppressor gene, p16INK4A. Deficient levels of MTAP can occur in cancers primarily through co-deletion of the MTAP gene and the p16INK4A gene.
