It specifically stains the nuclei of BT-20 cells and primary human foreskin fibroblast (HFF) cells. It does not stain the Rb-negative BT549 cells. It reacts with the hyperphosphorylated as well as the un (under) phosphorylated form of the Rb protein. Retinoblastoma gene product plays a key role in cell cycle control. Geminin is a nuclear protein that regulates the initiation of DNA replication during the cell cycle. DNA replication requires the coordinated association of Cdc6 and minichromosome maintenance (MCM) proteins with chromatin. Geminin blocks this assembly of the MCM into the prereplication complex and, in turn, prevents replication from occurring. Expression of Geminin fluctuates throughout the cell cycle with Geminin levels lowest at G1. Throughout S, G2 and M phases, Geminin levels are consistently elevated followed by a decrease during mitosis. The initiation of DNA replication is dependent on the degradation of Geminin during mitosis and the absence of Geminin throughout G1 phase. Recognizes a protein of 110 kDa, identified as Excision Repair Cross Complementing 1 (ERCC1). It is a mammalian nucleotide excision repair (NER) enzyme involved in repair of damaged DNA. ERCC1 is a homologous to RAD10 in Saccharomyces cerevisiae, which is required in mitotic intrachromosomal recombination and repair. ERCC1 is required in repair of cisplatin-induced DNA adducts and ultraviolet (UV)-induced DNA damage. This antibody is a mixture of four different monoclonal antibodies. This antibody is specific to 60 kDa latent membrane protein (LMP-1) encoded by the BNLF1 gene of the EBV. Each clone reacts with different epitopes on the hydrophilic C-terminus of the cytoplasmic domain of LMP-1. This antibody stains strongly with EBV-positive lymphoblastoid cell lines and EBV infected B cell immunoblasts in infectious mononucleosis. EBV, also designated human herpesvirus 4 (HHV-4), is a member of the herpesvirus family and is one of the most common human viruses. This antibody recognizes a protein of 40 kDa, identified as cytokeratin-19 (CK19), which is expressed in sweat gland, mammary gland ductal and secretory cells, bile ducts, gastrointestinal tract, bladder urothelium, oral epithelia, esophagus, and ectocervical epithelium. Anti-CK19 reacts with a wide variety of epithelial malignancies including adenocarcinomas of the colon, stomach, pancreas, biliary tract, liver, and breast. Perhaps the most useful application is the identification of thyroid carcinoma of the papillary type, although 50%-60% of follicular carcinomas are also labeled. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Epitope of this MAb maps in the N-terminus of PTH, a hormone produced by the parathyroid gland that regulates the concentration of calcium and phosphorus in extracellular fluid. This hormone elevates blood Ca2 levels by dissolving the salts in bone and preventing their renal excretion. It is produced in the parathyroid gland as an 84 amino acid single chain polypeptide. It can also be secreted as N-terminal truncated fragments or C-terminal fragments after intracellular degradation, as in case of hypercalcemia. Defects in this gene are a cause of familial isolated hypoparathyroidism (FIH); also called autosomal dominant hypoparathyroidism or autosomal dominant hypocalcemia.
