Calcineurin B homologous protein 2, also known as hepatocellular carcinoma-associated antigen 520, is a 196 amino acid protein that contains 4 EF-hand domains and plays a potential role in transmembrane Na/H exchange. By binding to and activating NHE-1, CHP2 increases the pH and works to protect cells from serum deprivation-induced death. Though not typically detected in normal tissues, CHP2 is highly expressed in malignantly transformed cells and is therefore considered to be a tumor-associated antigen. Ectopic expression of CHP2 promotes proliferation of HEK293 cells and knockdown of CHP2 mRNA in HepG2 cells inhibits cell proliferation. Like calcineurin B, CHP2 can bind to and stimulate phosphatase activity of calcineurin A and activate the calcineurin/NFAT signaling pathway. Arachidonic acid is converted by microsomal CYP450 enzymes to a variety of epoxides, ω-1 and ω-hydroxylated compounds via what is known as the epoxidase pathway.{313,403,823} MS-PPOH is a selective inhibitor of the epoxygenation reactions catalyzed by specific CYP450 isozymes.{8589} MS-PPOH inhibits the formation of arachidonate 11,12-epoxides by CYP4A2 and CYP4A3 enzymes with an IC50 value of 13 µM, but has no effect on the formation of 20-HETE, the ω-hydroxylation product of CYP4A1.{8588}

Rabbit IgG TrueBlot may also be used for detection in immunoblotting assays that do not employ immunoprecipitation. Rabbit IgG TrueBlot is provided as 1000X solution. We recommend incubating the blots from minigels in sealed bags (removing as much air as possible) with minimal volume (2-3 mls). If used conservatively at 2.5mls/blot will yield enough reagent for 80 blots. Note that there are three key procedural considerations: 1. Protein A or G should not be used for the IP. Use of protein A or G beads with the rabbit TrueBlot will result in contaminating bands. For IP, Anti-Rat IgG beads, or Anti-Rabbit IgG beads should be used for rat or rabbit immunoprecipitating antibodies, respectively. 2. Immunoprecipitate should be completely reduced. 3. BLOTTO/Milk should be used as the blocking protein for the immunoblot. Specific conditions for each protein and antibody combination should be specifically optimized by the end user.

If used conservatively at 2.5mls/blot will yield enough reagent for 40 blots. Note that there are three key procedural considerations: 1. Protein A or G beads may be used with the mouse, goat and sheep TrueBlot secondaries, but not with the rabbit TrueBlot secondary. 2. Immunoprecipitate should be completely reduced. 3. MB-070 Blocking Buffer for Fluorescent WB should be used as the blocking protein for the immunoblot. Note: To achieve best results when detecting mouse IgG1 subtypes, we recommend performing a dot blot or titration to determine the optimal dilution. Recognizes a protein of 55 kDa, which is identified as GLUT-1. Glucose transporters are integral membrane glycoproteins involved in transporting glucose into most cells. There are many types of glucose transport carrier proteins, designated as Glut-1 to Glut-12. Glut-1 is a major glucose transporter in the mammalian blood-brain barrier. It is expressed in high density on the membranes of human erythrocytes and the brain capillaries that comprise the blood-brain barrier. Glut-1 is expressed at variable levels in many human tissues. Overexpression of Glut-1 has been linked to tumor progression or poor survival of patients with carcinomas of the colon, breast, cervical, lung, bladder and mesothelioma. CD10 (also known as Common Acute Lymphocytic Leukemia Antigen (CALLA)) is a type II transmembrane protein. It is a cell surface enzyme with neutral metalloendopeptidase activity, which inactivates a variety of biologically active peptides. CD10 is expressed on the cells of lymphoblastic, Burkitt's, and follicular germinal center lymphomas, and on cells from patients with chronic myelocytic leukemia (CML). This antibody recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. Monoclonal antibodies (MAbs) to liver cell processes are useful in the identification of hepatic carcinomas and normal organ structures. MAb HSA98 binds to human hepatocytes and the majority of human hepatocellular carcinomas (HCC's). In frozen sections, it stains hepatic cells and may be used as a marker of the liver. Cell preparations of hepatocellular carcinoma biopsies or cell lines are found to bind HSA98 on the cell surface. This antibody recognizes a protein of 31 kDa, which is identified as MTAP (5'-deoxy-5'-methylthioadenosine phosphorylase). It catalyzes the reversible phosphorolysis of methylthioadenosine, which is important in polyamine metabolism and for the salvage of adenine and methionine. The gene encoding MTAP is linked to the tumor suppressor gene, p16INK4A. Deficient levels of MTAP can occur in cancers primarily through co-deletion of the MTAP gene and the p16INK4A gene. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms.
