m EI (Membrane Enterococcus Indoxyl) Agar is a chromogenic selective medium used for the detection and enumeration of Enterococcus species in recreational fresh and marine waters. The plates contain approximately 8 mL of agar and are cast in a 60 × 15 mm format designed to accommodate standard membrane filters without excess media. This formulation is the basis of EPA Method 1600 and enables a single step testing procedure. The medium contains the chromogenic substrate indoxyl β D glucoside, which is cleaved by β glucosidase enzymes produced by Enterococcus species. As a result, colonies develop a distinctive blue coloration or blue halo after incubation. Selective agents such as sodium azide and nalidixic acid inhibit competing bacteria, ensuring accurate detection of Enterococcus contamination in water samples.
