Biotium Human Kappa Light Chain(KLC264), CF488A conjugate, 0.1mg/mL
This MAb is specific to kappa light chain of immunoglobulin and shows no cross-reaction with lambda light chain or any of the five heavy chains. In mammals, the two light chains in an antibody are always identical, with only one type of light chain, kappa or lambda. The ratio of Kappa to Lambda is 70:30. However, with the occurrence of multiple myeloma or other B-cell malignancies this ratio is disturbed. Antibody to the kappa light chain is reportedly useful in the identification of leukemias, plasmacytomas, and certain non-Hodgkin's lymphomas. This antibody recognizes a protein of ~35 kDa, identified as CD74 (Workshop IV). CD74 is a type II transmembrane protein which binds to the peptide binding groove of newly synthesized MHC class II alpha/beta heterodimers and prevents their premature association with endogenous polypeptides. CD74 is expressed primarily by antigen presenting cells, such as B-lymphocytes (from before the pre-B cell stage to before the plasma cell stage), macrophages, and monocytes, and many epithelial cells. Anti-CD74 stains predominantly germinal center lymphocytes and B-cell lymphomas, but rarely T-cell lymphomas. The onset of angiogenesis is believed to be an early event in tumorigenesis and may facilitate tumor progression and metastasis. Several growth factors with angiogenic activity have been described. These include Fibroblast Growth Factor (FGF), Platelet Derived Growth Factor (PDGF), Vascular Endothelial Growth Factor (VEGF) and Placenta Growth Factor (PLGF). Placenta growth factor (PLGF) is a secreted protein primarily produced by placental trophoblasts but also expressed in other endothelial cells and tumors. There are three isoforms, PLGF-1, PLGF-2, and PLGF-3. PLGF-2 is expressed up until week 8 in the placenta; the placental tissues continuously express PLGF-1 and PLGF-3 but only PLGF-1 is found in colon and mammary carcinomas. This MAb recognizes a 21 kDa protein, identified as the p21WAF1 tumor suppressor protein. This MAb is highly specific to p21 and shows no cross-reaction with other closely related mitotic inhibitors. p21WAF1 is a specific inhibitor of cdk s and a tumor suppressor involved in the pathogenesis of a variety of malignancies. The expression of this gene acts as an inhibitor of the cell cycle during G1 phase and is tightly controlled by the tumor suppressor protein p53. Its expression is induced by the wild type, but not mutant, p53 suppressor protein. Normal cells generally display a rather intense nuclear p21 expression. Loss of p21 expression has been reported in many carcinomas (gastric carcinoma, non-small cell lung carcinoma, thyroid carcinoma). CD11c, also known as Integrin, alpha X (complement component 3 receptor 4 subunit) (ITGAX), is a type I transmembrane protein found at high levels on most human dendritic cells, but also on monocytes, macrophages, neutrophils, and some B cells. It induces cellular activation and helps trigger neutrophil respiratory burst. AM1-43 is a fixable, activity-dependent fluorescent nerve terminal probe. AM1-43 functions similarly similarly to SynaptoGreen™ C4, but with the additional property of being fixable. Thus, AM1-43 is a useful tool for synapse studies where subsequent fluorescent immunochemistry is desired. AM1-43 and FM1-43 have identical excitation and emission wavelengths. Biotium p40(Rabbit PAb), CF568 conjugate, 0.1mg/mL
p40 (p63 delta) is a marker determined to be highly specific for squamous basal cells by immunohistochemistry (IHC). The current more routinely recommended marker, p63, appears to have less specificity compared to p40, especially on squamous cell tumors. The ability to differentiate between lung adenocarcinoma vs. squamous cell carcinoma is difficult and has bearing on the different therapeutic avenues for each subtype treatment. p63 antibody's ability to distinguish between the tumor types appears to be inferior when compared to p40. CD11b is a cell adhesion molecule that acts as a receptor for cell surface ligands such as intracellular adhesion molecules (ICAMs) or soluble ligands. Integrins are heterodimeric proteins that contain an alpha chain and beta chain. IntegrinαM combines with the Integrinβ2 to form a leukocyte-specific integrin referred to as macrophage receptor 1 (Mac-1), or inactivated-C3b (iC3b) receptor 3 (CR3). IntegrinαM/β2 is important in the adherence of neutrophils and monocytes to stimulated endothelium, and also in the phagocytosis of complement coated particles. The protein CD11b has been implicated in the various adhesion-related interactions of cells such as monocytes, macrophages, natural killer (NK) cells, and granulocytes. It is part of a heterodimer that consists of CD11b and CD18. It also modulates the uptake of complement-coated particles within the cell. Recognizes a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesized as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkin's disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. It is also expressed by the tumor cells of a majority of anaplastic large cell lymphomas as well as by a varying proportion of activated T and B cells. This MAb distinguishes large cell lymphomas derived from activated lymphoid cells from histiocytic malignancies and lymphomas derived from resting and precursor lymphoid cells or from anaplastic carcinomas. Recognizes a protein of a disulfide-linked 120 kDa dimer, identified as CD27 (Workshop VI; Code 6T-028). CD27 is expressed on the majority of peripheral T cells, medullary thymocytes, memory-type B cells, and natural killer cells. It is a transmembrane phosphoglycoprotein that belongs to the tumor necrosis factor receptor (TNFR) superfamily. CD27 binds to its ligand CD70, a member of the TNF family, and induces T-cell co-stimulation and B-cell activation.
