Metal enhanced DAB technology permits detection of extremely low concetrations of tissue antigens. This kit provides a sensitive means for detecting peroxidase labeled antibodies and conjugates. HistoMark Orange contains nickel enhanced DAB that produces an orange-red DAB polymer and also exceeds the sensitivity of DAB alone. The kit contains the nuclear counterstain, Contrast GREEN, a solution of methyl green that contrasts sharply with orange. Blocking solution is provided to prevent endogenous peroxidase activity. The precipitates from HistoMark Orange are alcohol and xylene insoluble, allowing section to be mounted in aqueous or xylene-based mounting media.

The gamma globin genes (HBG1 and HBG2) are normally expressed in the fetal liver, spleen and bone marrow. Two gamma chains together with two alpha chains constitute fetal hemoglobin (HbF) which is normally replaced by adult hemoglobin (HbA) at birth. In some beta-thalassemias and related conditions, gamma chain production continues into adulthood. The two types of gamma chains differ at residue 136 where glycine is found in the G-gamma product (HBG2) and alanine is found in the A-gamma product (HBG1). The former is predominant at birth. The order of the genes in the beta-globin cluster is 5-epsilon -- gamma-G -- gamma-A -- delta -- beta--3.

3, 3’ Diaminobenzidine (DAB) is a widely used chromogen for immunohistochemical staining and immunoblotting. When in the presence of peroxidase enzyme, DAB produces a brown precipitate that is insoluble in alcohol and xylene. This product comes in a two-component system consisting of a liquid stable DAB chromogen and DAB substrate buffer. The color of the chromogen may vary from pale brown to orange. However, this does not interfere with staining properties. intelliPATH FLX DAB is a very stable and superior formulation. In some cases, antibody titers may increase by two-fold. intelliPATH FLX DAB can be used both manually and on automated stainers.

Pronase is used for proteolytic digestion of formalin-fixed paraffin-embedded (FFPE) tissue sections prior to application of antibodies. In IHC most commonly used fixative like formalin mask tissue antigens (cellular, membrane and nuclear) by cross-linking process, this results in poor or no staining in IHC. Pronase digestion of FFPE tissue section improves accessibility of antibodies to tissue antigens.

Intented Use: Use as antigen retriever for some antibodies in IHC. Please refer to primary antibody protocol for IHC.

This antibody reacts with a carbohydrate determinant, identified as CDw78 (Workshop V; Code CDw78.2). Epitope recognized by this MAb and Leu21 is the same. It is present on some immature and some peripheral blood B cells, and some B cell lines. Its expression is increased after B-cell activation in vitro. CDw78 is also found on tissue macrophages and on epithelial cells, but not on T cells, NK cells, monocytes, granulocytes and myeloid tissues. It appears that CDw78 is a determinant on a major histocompatibility complex (MHC) class II subpopulation.

Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). 3-D Life is the most flexible system for biomimetic hydrogel design for 3D cell cultures. It uses two polymers that form a hydrogel by covalent crosslinking at ambient temperature. The chemical reaction is non-toxic to cells and occurs rapidly, preventing cell settlement before gel is formed. The 3-D Life ToGro Ready-To-Go Hydrogel Kit allows preparation of hydrogels with soft stiffness (400 Pa; shear modulus) and 0.4 mmol/L RGD Peptide concentration. This gel allows 3D spreading and migration of most cells. The gel is formed by crosslinking of RGD-Dextran with CD-Link. Thiol groups on CD-Link form stable thioether bonds with thiol-reactive groups on RGD-Dextran resulting in gel formation within 20 min. CD-Link is designed for MMP cleavage, allowing cell spreading/migration if they express the indicated MMPs. ToGro hydrogels can be dissolved by adding dextranase, allowing easy cell recovery.
