Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.Immunohistochemical staining with this MAb is indistinguishable from that obtained with monoclonal antibody 5D3.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. The protein encoded by this gene is a member of the transmembrane 4 superfamily, also known as the tetraspanin family. Most of these members are cell-surface proteins that are characterized by the presence of four hydrophobic domains. The proteins mediate signal transduction events that play a role in the regulation of cell development, activation, growth and motility. This encoded protein is found in the asymmetrical unit membrane (AUM) where it can complex with other transmembrane 4 superfamily proteins. It may play a role in normal bladder epithelial physiology, possibly in regulating membrane permeability of superficial umbrella cells or in stabilizing the apical membrane through AUM/cytoskeletal interactions. 3-D Life is the most flexible system for biomimetic hydrogel design for 3D cell cultures. It uses two polymers that form a hydrogel by covalent crosslinking at ambient temperature. The chemical reaction is non-toxic to cells and occurs rapidly, preventing cell settlement before gel is formed. The 3-D Life ToGro Ready-To-Go Hydrogel Kit allows preparation of hydrogels with soft stiffness (400 Pa; shear modulus) and 0.4 mmol/L RGD Peptide concentration. This gel allows 3D spreading and migration of most cells. The gel is formed by crosslinking of RGD-Dextran with CD-Link. Thiol groups on CD-Link form stable thioether bonds with thiol-reactive groups on RGD-Dextran resulting in gel formation within 20 min. CD-Link is designed for MMP cleavage, allowing cell spreading/migration if they express the indicated MMPs. ToGro hydrogels can be dissolved by adding dextranase, allowing easy cell recovery.
