The conjugated goat anti-mouse polymer horseradish peroxidase (HRP) and the conjugated goat anti-rabbit polymer alkaline phosphatase (AP) secondary antibodies react with both heavy and light chains on mouse and rabbit IgG respectively. This innovative HRP- and AP-polymerization technology provides a significant increase in staining sensitivity when compared to other conventional HRP- or AP-conjugated secondary antibodies. Avidin-biotin blocking procedures are not necessary when using the MACH 2 conjugated secondary antibodies. It is specially designed for a rapid double stain procedure. An antibody cocktail with a mouse monoclonal and a rabbit polyclonal/monoclonal must be used. The overall staining procedure can be done with 4 major steps and in less than two hours. It can be used manually or used with automated stainers.

Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.Immuno-histochemical staining with this MAb is indistinguishable from that obtained with monoclonal antibody 5D3.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). CK8 is primarily found in the non-squamous epithelia and is present in majority of adenocarcinomas and ductal carcinomas. It is absent in squamous cell carcinomas. Hepatocellular carcinomas are defined by the use of antibodies that recognize only cytokeratin 8 and 18. CK8 exists on several types of normal and neoplastic epithelia, including many ductal and glandular epithelia such as colon, stomach, small intestine, trachea, and esophagus as well as in transitional epithelium. Anti-CK8 does not react with skeletal muscle or nerve cells. Epithelioid sarcoma, chordoma, and adamantinoma show strong positivity corresponding to that of simple epithelia (with antibodies against CK8, CK18 and CK19). Reportedly, anti-CK8 is useful for the differentiation of lobular ('ring-like, perinuclear') from ductal ('peripheral-predominant')

S100 belongs to the family of calcium binding proteins. S100A and S100B proteins are two members of the S100 family. S100A is composed of an alpha and a beta chain whereas S100B is composed of two beta chains. This antibody is specific against an epitope located on the beta-chain (i.e. in S-100A and S-100B) but not on the alpha-chain of S-100 (i.e. in S-100A and S100A0). The protein encoded by this gene is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This kinase is activated by various environmental stresses and proinflammatory cytokines. The activation requires its phosphorylation by MAP kinase kinases (MKKs), or its autophosphorylation triggered by the interaction of MAP3K7IP1/TAB1 protein with this kinase.
