Cayman’s PC Assay provides a specific, sensitive, and convenient method for quantifying PC in plasma or serum. In this assay, PC-Specific PLD hydrolyzes PC to choline and phosphatidic acid. The newly formed choline is then used to generate hydrogen peroxide in a reaction catalyzed by choline oxidase. Finally, with peroxidase as a catalyst, hydrogen peroxide reacts with DAOS and 4-aminoantipyrine to generate a blue dye with an optimal absorption at 595 nm.
Cayman Chemical Triglyceride Colorimetric Ass
Cayman’s Triglyceride Colorimetric Assay Kit provides a simple, reproducible, and sensitive tool for assaying mono-, di-, and triglycerides in plasma, serum, cell culture extracts, and tissue homogenates. The Triglyceride Colorimetric Assay Kit uses the enzymatic hydrolysis of triglycerides by lipase to produce glycerol and free fatty acids. The glycerol released is subsequently measured by a coupled enzymatic reaction system. The glycerol formed in reaction 1 is phosphorylated to glycerol-3-phosphate in a reaction catalyzed by glycerol kinase. The glycerol-3-phosphate is oxidized by glycerol phosphate oxidase producing dihydroxyacetone phosphate and hydrogen peroxide (H2O2). Peroxidase catalyzes the redox-coupled reaction of H2O2 with 4-aminoantipyrine (4-AAP) and N-ethyl-N-(3-sulfopropyl)-m-anisidine (ESPA), producing a brilliant purple color. The absorbance is measured at 540 nm.

Cayman’s Protein Carbonyl Colorimetric Assay Kit utilizes the DNPH reaction to measure the protein carbonyl content in plasma, serum, cell lysates, or tissue homogenates in a convenient 96-well format. The amount of protein-hydrazone produced is quantified spectrophotometrically at an absorbance between360-385 nm. The carbonyl content can then be standardized to the protein concentration.

Glycerol is the backbone of triglycerides and an important intermediate in energy metabolism being involved in both oxidative and synthetic processes. The circulating levels of glycerol and free fatty acids are considered to reflect lipolysis and are therefore useful parameters to evaluate in various conditions. The measurement of glycerol is also useful for correction of glycerol interference when measuring triglycerides in reference materials and in serum from patients with elevated glycerol concentrations. Fluo-Forte Calcium Assay Kits detect mobilization of intracellular calcium utilizing a fluorogenic calcium-binding dye optimized for superior cell-permeability and retention. The self-quenching dye undergoes an electronic change upon binding of calcium, resulting in a several order of magnitude greater fluorescence. Mechanism of Action The fluorogenic calcium-binding dye is provided to the cells as an acetoxymethyl (AM) ester, which is cell-permeable. Once inside the cells, the dye is hydrolyzed by intracellular esterases, which leads to generation of a cell membrane impermeable negatively charged form. In the absence of calcium, the calcium-binding moiety portion of the probe quenches the fluorescence of the fluorophore portion of the probe by photo-induced electron transfer. Cayman’s Uric Acid Assay Kit provides fluorescence- or colorimetric-based methods for detecting uric acid in plasma, serum, and urine. In the assay, uricase catalyzes the conversion of uric acid to allantoin, hydrogen peroxide (H2O2), and carbon dioxide. H2O2, in the presence of horseradish peroxidase, reacts stoichiometrically with ADHP (10-acetyl-3,7-dihydroxyphenoxazine) to produce the highly fluorescent compound resorufin. Resorufin fluorescence can be analyzed with an excitation wavelength between 530-540 nm and an emission wavelength between 585-595 nm. Alternatively, the absorbance of resorufin can be measured at 570 nm.

Cayman’s Superoxide Dismutase Assay Kit provides a convenient method of measuring SOD activity in plasma, serum, erythrocyte lysates, tissue homogenates, and cell lysates. The assay relies on xanthine oxidase to produce superoxide radicals, which then reduce a tetrazolium salt to a formazan dye, detectable at 450 nm. SOD in the sample inhibits this reduction by catalyzing the dismutation of superoxide radicals. Thus, the degree of inhibition is directly proportional to SOD activity. The SOD Assay Kit measures all three types of SOD (Cu/Zn-, Mn-, and Fe-SOD). Mitochondrial Mn-SOD can be assayed separately following the procedure outlined in the kit booklet.
