The InCELL Hunter eXpress MEK1 Kinase Binding Assay provides an easy-to-use cell based assay to study small molecule drug candidates that engage with kinases. This kit assesses compound-engagement-based stabilization of MEK1 via detection of MEK1 levels. This product uses EFC technology. The cell line included overexpresses MEK1 target protein fused to ProLabel. In the absence of compound that binds and stabilizes the target, this protein is degraded and no EFC signal is produced. When compound engages and stabilizes the target, EA contained in the detection reagent produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Cayman’s Hydrogen Peroxide Assay Kit provides fluorometric or colorimetric methods for the quantification of extracellular H2O2 produced by cultured cells. In the presence of horseradish peroxidase (HRP), H2O2 reacts stoichiometrically with an HRP substrate, ADHP, to produce the compound resorufin. Resorufin fluorescence can be easily quantified at excitation and emission wavelengths of 530-540 and 585-595 nm, respectively. Alternatively, the absorbance of resorufin can be measured at 570 nm. Catalase, an H2O2 scavenger, is included in the kit for verifying the specificity of the assay.
