The InCELL Hunter eXpress HEK 293 BRD2(1) Bromodomain Assay provides an easy-to-use cell based assay to study small molecule drug candidates that engage with regulators of epigenetics pathways. This kit assesses compound-engagement-based stabilization of BRD2(1) via detection of BRD2(1) levels. This product uses EFC technology. The cell line included overexpresses BRD2(1) target protein fused to ProLabel. In the absence of compound that binds and stabilizes the target, this protein is degraded and no EFC signal is produced. When compound engages and stabilizes the target, EA contained in the detection reagent produces EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. This kit assesses ligand (e.g. CP 55,940) based activation of CNR2 GPCR activity via detection of decreasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type CNR2, a naturally Gi/Go coupled GPCR. Upon activation of CNR2, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates.
