The cAMP Hunter MC3R eXpress kit provides an easy-to-use cell based assay to study drug candidates (small molecules or biologics) targeting GPCRs. This kit assesses ligand (e.g. α-melanocyte stimulating hormone) based activation of MC3R GPCR activity via detection of increasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type MC3R, a naturally Gs coupled GPCR. Upon activation of MC3R, the increase in levels of intracellular cAMP leads to an increased signal from EFC when the cAMP detection kit is applied. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Adhesion to the extracellular matrix is essential for the survival of many adherent cells. Apoptosis that results from the loss of cell adhesion to the ECM, or inappropriate adhesion, is defined as “anoikis”. A common feature of carcinoma development is the ability of transformed cells to survive under anchorage independent or “spheroid” growth conditions. Resistance to anoikis has been shown to be involved in the loss of cell homeostasis, cancer growth, and metastasis. Preventing anoikis is a major goal in the development of cell transplantation techniques, including the therapeutic use of progenitor cells. The CytoSelect™ 96-well Anoikis Assay Kit provides a colorimetric and fluorometric format to measure anchorage-independent growth and monitoring anoikis propelled cell death. The kit contains sufficient reagents for 96 samples in a hydrogel-coated 96-well plate. Live cells are detected with MTT or Calcein AM. Cell death is detected with the Ethidium Homodimer (EthD-1).
