This kit uses EFC technology to measure ligand-induced internalization of CMKLR1 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged CMKLR1. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting CMKLR1-arrestin complex is internalized to the early endosome, leading to production of EFC signal. eXpress kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, detection reagent, and assay plates. Cayman’s HDAC Cell-Based Assay Kit provides a robust and EAsy-to-use platform for studying HDAC activity modulators in whole cells. By using a cell-permeable HDAC substrate, the activity of various lysine-specific protein deacetylases can be measured in intact cells in a simple and homogenous manner. The fluorescence of the deacetylated reaction product can be analyzed using a fluorometer with an excitation wavelength of 340-360 nm and an emission wavelength of 440-460 nm.
