Tribioscience’s Tyrosinase Activity Assay Kit provides a simple, sensitive assay for the measurement of tyrosinase activity in biological samples. In this assay, tyrosinase catalyzes the conversion of a phenolic substrate to a Quinone intermediate, which reacts with a probe forming a highly stable chromophore with absorbance at 510 nm. The assay can detect as low as 30 µU Tyrosinase in biological samples. The kit is enough for 100 assays.

Cayman Chemical HDAC Fluorometric Activity As
Cayman’s HDAC Fluorometric Activity Assay Kit provides a fast, fluorometric method for measuring HDAC activity without the use of radioactivity, extraction, or chromatography. In the first step, an acetylated lysine substrate is incubated with samples. Deacetylation sensitizes the substrate such that treatment with the HDAC developer in the second step releases a fluorescent product. Trichostatin A is included as a control inhibitor. The fluorophore can be EAsily analyzed using a fluorescence plate reader with an excitation wavelength of 340-360 nm and an emission wavelength of 440-465 nm. The assay can be used for quantifying Class I and II HDAC activity from various sources.

Cayman’s L-Lactate Assay provides both a fluorescence and an absorbance-based method for detecting L-lactate in biological samples such as serum, plasma, blood, urine, and saliva. It can also be utilized to determine intracellular and extracellular lactate concentrations in cell culture samples. In the assay, LDH catalyzes the oxidation of lactate to pyruvate, along with the concomitant reduction of NAD+ to NADH. NADH reacts with the substrate to yield a final product that can be measured fluorometrically with an excitation wavelength of 530-540 nm and an emission wavelength of 585-595 nm, or colorimetrically at 535 nm.

Cayman’s cGAS Inhibitor Screening Assay Kit provides a robust and EAsy-to-use platform for identifying novel inhibitors of human cGAS, which is a cytosolic dsDNA sensor that detects microbial pathogens. The assay directly measures 2’3’-cGAMP produced by cGAS in the presence of DNA, ATP, and GTP. The cyclic dinucleotide product of that reaction is quantified via ELISA using a 2’3’-cGAMP-specific antiserum. The cGAS inhibitor CU-76 is included as a positive control. The ELISA portion of Cayman’s cGAS Inhibitor Screening Assay Kit is a competitive assay with a range of 4.57-10,000 pM, a midpoint of approximately 300 pM (50% B/B0), and a sensitivity (80% B/B0) of approximately 50 pM.

Hemoglobin (Hb or Hgb) is a globular protein found primarily in erythrocytes that carries oxygen from the lungs to tissue where it releases oxygen and then returns carbon dioxide (CO2) from the tissue to the lungs. Hemoglobin also binds CO2, carbon monoxide (CO), and nitric oxide (NO). Cayman’s Hemoglobin Colorimetric Assay provides a quick, reliable method for determining total hemoglobin concentration in a variety of biological samples, including blood, tissue homogenates and cell lysates. Unlike the universally accepted reference method of hemoglobin determination, which uses potassium cyanide as a reagent and commonly under-estimates hemoglobin levels, Cayman’s Hemoglobin Colorimetric Assay utilizes an optimized detection reagent which is non-toxic, reporting accurate measurements of total hemoglobin concentrations with an absorbance between 560-590 nm.

Cayman’s Nitrate/Nitrite Colorimetric Assay Kit provides an accurate and convenient method for measurement of total and individual nitrate and nitrite concentrations in a simple two-step process utilizing nitrate and nitrite standards. The first step is the conversion of nitrate to nitrite utilizing nitrate reductase. The second step is the addition of the Griess Reagents which convert nitrite into a deep purple azo compound (See Figure 2).{1191} Photometric measurement of the absorbance due to this azo chromophore accurately determines NO2- concentration. This kit can be used to measure nitrate and nitrite in plasma/serum, urine, tissue culture media, and tissue homogenates. However, it cannot be used to analyze nitrate and nitrite from an in vitro assay of nitric oxide synthase (NOS) in which excess NADPH has been added. For these assays a second method (LDH method) is utilized and is available from Cayman Chemical in a 96 well plate format (Item No. 760871).
