A DNA sequence encoding the Human SLC39A10 (Q9ULF5)(Leu338-Phe831) was expressed with a polyhistidine tag at the C-terminus. This protein was prepared using our Nanodisc platform. Nanodisc is a versatile tool for studying membrane proteins. Using styrene-maleic acid (SMA) copolymer, membrane proteins can be extracted directly from prokaryotic and eukaryotic expression systems in the absence of detergents to preserve the protein structure and function better. Compared to membrane scaffold proteins (MSPs) nanodiscs, SMA nanodiscs also have the advantage of preserving proteins' nature by maintaining native lipids surrounded without introducing any heterologous proteins, which allows studies of protein structure and functions in a native-like environment.

The protein encoded by this gene is a member of the S100 family of proteins containing 2 EF-hand calcium-binding motifs. S100 proteins are localized in the cytoplasm and/or nucleus of a wide range of cells, and involved in the regulation of a number of cellular processes such as cell cycle progression and differentiation. S100 genes include at least 13 members which are located as a cluster on chromosome 1q21, however, this gene is located at 21q22.3. This protein may function in Neurite extension, proliferation of melanoma cells, stimulation of Ca2+ fluxes, inhibition of PKC-mediated phosphorylation, astrocytosis and axonal proliferation, and inhibition of microtubule assembly. Chromosomal rearrangements and altered expression of this gene have been implicated in several neurological, neoplastic, and other types of diseases, including Alzheimers disease, Downs syndrome, epilepsy, amyotrophic lateral sclerosis, melanoma, and type I diabetes.

Members of arrestin/beta-arrestin protein family are thought to participate in agonist-mediated desensitization of G-protein-coupled receptors and cause specific dampening of cellular responses to stimuli such as hormones, neurotransmitters, or sensory signals. Arrestin beta 2, like arrestin beta 1, was shown to inhibit beta-adrenergic receptor function in vitro. It is expressed at high levels in the central nervous system and may play a role in the regulation of synaptic receptors. Besides the brain, a cDNA for arrestin beta 2 was isolated from thyroid gland, and thus it may also be involved in hormone-specific desensitization of TSH receptors. Multiple alternatively spliced transcript variants encoding different isoforms have been found for this gene.
