S2Media™ Spot Indole Reagent is used to determine the ability of microorganisms to split indole from the amino acid tryptophan. This reaction is mediated by the enzyme tryptophanase, which hydrolyzes tryptophan to produce indole. Indole then reacts with p-dimethylaminocinnamaldehyde (DMACA) to form a blue-green compound. Testing is performed on isolated colonies on solid media.Positive: Blue or blue-green color (indole production).Negative: Pink or no color change (no indole production).The spot test provides a rapid method for differentiating indole-positive organisms (e.g., Escherichia coli) from indole-negative species.• Spot Indole Reagent should be used by trained professionals within a laboratory setting. • Product is not intended for use in the diagnosis or treatment of disease or other human conditions.
