25mM magnesium is for optimization of magnesium concentrations in PCR reactions. Optimal magnesium concentration increases fidelity and specificity of the polymerase. Too low of a concentration inactivates the polymerase while too high of a concentration increase nonspecific bands. Several variables effect the optimal concentration of magnesium, DNA quality, presence of chelators, and dNTP concentration.
