The 26S proteasome is the main proteasome responsible for ubiquitin‑mediated protein degradation and is essential for cellular homeostasis. It regulates the turnover of key proteins involved in apoptosis, the cell cycle, and signal transduction, and maintains protein quality by removing misfolded proteins from the cytosol and ER. Because of its central role, the proteasome is a major drug target, especially in cancer and neurodegeneration. Clinically approved inhibitors such as bortezomib and carfilzomib highlight its therapeutic importance, and additional inhibitors are in trials for multiple myeloma and other malignancies. Structurally, the 26S proteasome consists of the catalytic 20S core particle and one or two 19S regulatory particles, which mediate deubiquitylation, unfolding, and substrate delivery into the 20S chamber.
