3-Methyl-2-buten-1-ol is an orally active endogenous metabolite found in pine trees, utilized as a fragrance ingredient in cosmetics and detergents. This compound can pass the cell barrier and has been observed to reduce body weight in rats. In vitro studies show that it quickly passes the Caco-2 cell barrier with a Papp of 37.3 μcm/s. In vivo, concentrations of 200-5000 ppm in drinking water over 90 days lead to decreased food and water consumption, body weight, food efficiency, and urinary volume in rats. Additionally, doses of 50-600 mg/kg via gavage from days 6-19 post coitum resulted in unscheduled death of one dam and transient signs like salivation, abdominal position, lacrimation, and/or piloerection in rats.
