7-AAD Cell Viability Assay Kit utilizes the fluorescent nucleic acid dye 7-aminoactinomycin D (7-AAD) to differentiate viable cells from apoptotic or necrotic cells 7-AAD is membrane-impermeable and specifically labels nucleic acids in cells with compromised membrane integrity a common feature of dead or damaged cells Upon binding DNA within such cells 7-AAD emits fluorescence detectable under excitation near 488 nm and emission around 647 nm This dye is frequently applied in conjunction with Annexin V-FITC or calcein AM in apoptosis necrosis and cell viability assays or employed to exclude dead cells in flow cytometry analysis The recommended staining concentration is typically 5 g/mL to clearly distinguish viable and compromised cells detectable by fluorescence microscopy or flow cytometry
