This gene encodes a member of the C1 family of peptidases. Alternative splicing results in multiple transcript variants, with at least one encoding a preproprotein that is processed into cathepsin B light and heavy chains, which dimerize to form the active enzyme. Cathepsin B is a lysosomal cysteine protease with endopeptidase and exopeptidase activity, involved in protein turnover. It also acts as amyloid precursor protein secretase, with incomplete APP processing linked to Alzheimers disease. Overexpression of this protein is associated with esophageal adenocarcinoma and other cancers. Both cathepsin B and L are involved in cleaving the spike protein of SARS-CoV-2 during cell entry. Multiple pseudogenes of this gene have been identified.
