Inhibit ALK4, ALK5, and ALK7 with SB431542 (Inman et al. Mol Pharmacol, 2002; Laping et al. Mol Pharmacol, 2002). This TGF-β/Activin/NODAL pathway inhibitor enhances the differentiation of cardiomyocytes from mouse and human PSCs (Kattman et al. Cell Stem Cell, 2011). In combination with various small molecules, SB431542 is used to increase the efficiency of reprogramming human somatic cells to iPS cells (Lin et al. Nat Methods, 2009), promote differentiation of neural progenitor cells from human PSCs (Chambers et al. Nat Biotechnol, 2009; Chambers et al. Nat Biotechnol, 2012), and modulate the direct lineage reprogramming of fibroblasts to mature neurons (Li et al. Cell Stem Cell, 2015).
