ACY-241 (CAS 1316215-12-9) is a selective orally bioavailable inhibitor of histone deacetylase 6 (HDAC6) an enzyme that removes acetyl groups from -N-acetyl lysine residues on histones thus modulating chromatin structure and gene expression In preclinical studies ACY-241 demonstrated synergistic induction of apoptosis in multiple myeloma and mantle cell lymphoma cells when combined with immunomodulatory agents such as lenalidomide or pomalidomide accompanied by reduced MYC and IRF4 expression In solid tumor models co-administration with paclitaxel enhanced mitotic defects and led to dose-dependent hyperacetylation of -tubulin These findings support ACY-241 as a tool compound for investigating HDAC6 modulation in hematological and solid malignancies
