Triple Sugar Iron (TSI) Agar is a differential culture medium used for the identification of Enterobacteriaceae and other Gram-negative bacteria. The medium contains three sugars (glucose, sucrose, and lactose) and iron salts, which are essential for detecting bacterial fermentation and hydrogen sulfide production. The pH indicator phenol red changes color in response to acid production, while the iron salt reacts with hydrogen sulfide to form a black precipitate. The slant/butt reaction helps determine whether bacteria ferment sugars and produce gas or hydrogen sulfide. This medium is widely used in diagnostic microbiology to differentiate between pathogens based on metabolic activity.
