Anagrelide hydrochloride is a potent inhibitor of phosphodiesterase type III (PDE3) with an IC50 of 36 nM. This imidazoquinazoline derivative inhibits platelet aggregation and bone marrow megakaryocytopoiesis. It also decreases gastrointestinal stromal tumor (GIST) cell proliferation and promotes their apoptosis in vitro, acting as a platelet-lowering and antithrombopoietic agent.
- Potently inhibits the development of marrow megakaryocytes (IC50 = 26 nM).
- Inhibits only megakaryocytic cell growth, not non-megakaryocytic cells, at concentrations of 0.05, 0.3, and 1 μM over 12 days.
- Induces a cytotoxic effect in the GIST882 cell line (IC50 = 16 nM) at concentrations ranging from 0.1 to 10000 nM.
- Inhibited or reduced tumor growth in GIST2B, GIST9, and GIST882 models in adult female athymic mice at a dosage of 5 mg/kg administered twice daily for 10 days.
