Tandem Ubiquitin Binding Entities (TUBEs) have become essential tools for studying polyubiquitin. They bind K48‑ and K63‑linked tetra‑ubiquitin with nanomolar affinity—100 to 1000 times stronger than monomeric UBAs—while protecting ubiquitinated proteins from DUBs and the proteasome. This enables efficient isolation of native polyubiquitin chains from cells and tissues under gentle conditions. TUBE 1 and TUBE 2 enrich all linkage types, making them useful when the linkage is unknown. K48 TUBE HF provides strong selectivity for K48 chains and can be combined with K63 or M1 TUBEs to analyze chain architecture. Overall, TUBEs offer a powerful platform for isolating, enriching, and characterizing polyubiquitinated proteins.
