The HK5.3 monoclonal antibody targets mouse ICOSL (inducible T cell co-stimulator ligand), also known as CD275, B7RP-1, and B7-H2. ICOSL is a 40 kDa immune checkpoint protein of the Ig receptor superfamily. Its binding co-stimulates T and B cell responses. It is expressed on antigen-presenting cells such as splenic B cells, dendritic cells, and macrophages. The HK5.3 antibody blocks ICOSL binding to ICOS both in vitro and in vivo, and its treatment in mice has been reported to lead to a loss of regulatory T cells.
