Annexin V is a Ca2 -dependent phospholipid binding protein that specifically binds to phosphatidylserine (PS) PS is normally located on the inner leaflet of the cell membrane lipid bilayer but during early apoptosis PS is translocated from the inner leaflet to the outer surface of the cell membrane exposed to the extracellular environment Annexin V has a high affinity for PS and when labeled with fluorophore (HF555) Annexin V can serve as a fluorescent probe to detect apoptosis via flow cytometry or fluorescence microscopy The staining process requires only 10-20 minutes SYTOX Green specifically binds to double-stranded DNA emitting green fluorescence and normally cannot cross intact cell membranes However in late-stage apoptotic cells or dead cells SYTOX Green can penetrate the cell membrane and stain the nucleus By combining Annexin V-HF555 and SYTOX Green staining it is possible to distinguish among early apoptotic late apoptotic and dead cells
