This gene encodes a member of the AKT serine-threonine kinase family (protein kinase B alpha, beta, and gamma). These proteins, which share a pleckstrin homology domain, a kinase domain, and a regulatory domain, are phosphorylated by phosphoinositide 3-kinase (PI3K). AKT/PI3K plays a key role in signaling pathways involving receptor tyrosine kinases, G-protein coupled receptors, and integrin-linked kinase, regulating functions like cell proliferation, survival, metabolism, and angiogenesis. Full activation of AKT1 requires phosphorylation at threonine 308 and serine 473. AKT1 is involved in apoptosis suppression, tumor cell survival, and the mTOR pathway, which regulates protein synthesis. Mutations in this gene are linked to various cancers and conditions like Proteus syndrome and Cowden syndrome. Multiple alternatively spliced transcript variants exist.
