Myosin phosphatase target subunit 1 (MBS) is a subunit of myosin phosphatase, which regulates the actin-myosin interaction downstream of the GTPase Rho. RhoA, in its active GTP-bound form, interacts with MBS to control myosin light chain (MLC) phosphorylation, affecting smooth muscle contraction and nonmuscle cell function. RhoA activates Rho-kinase, which phosphorylates MBS and inactivates myosin phosphatase. Overexpression or activation of RhoA increases MBS and MLC phosphorylation. This pathway regulates the balance of actin and myosin interaction. Several isoforms of this gene have been identified due to alternative splicing.
