This gene encodes one of three AKT serine-threonine kinases (AKT1, AKT2, AKT3), essential in regulating various cellular functions such as proliferation, survival, metabolism, and angiogenesis. AKT proteins have a pleckstrin homology domain, a kinase domain, and a regulatory domain. They are activated by phosphorylation through PI3K and are recruited to the cell membrane by PIP3. Activation requires phosphorylation at threonine 308 and serine 473, with additional phosphorylation in response to growth factors like IGF-1 and EGF. AKT1 is a key component in the PI3K/AKT signaling pathway, regulating tumor cell survival and apoptosis. AKT also participates in the mTOR pathway, which is crucial for protein synthesis. Mutations in this gene are linked to cancers (e.g., breast, ovarian, colorectal) and disorders like Proteus and Cowden syndromes. Alternative splicing produces different isoforms of this gene.
