Azido-PEG4-alcohol is a PEG-based PROTAC linker that can be used in the synthesis of PROTACs. It is a click chemistry reagent containing an Azide group, able to undergo copper-catalyzed azide-alkyne cycloaddition reaction (CuAAc) with Alkyne groups, and strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with DBCO or BCN groups. PROTACs themselves contain two different ligands connected by a linker; one for an E3 ubiquitin ligase and the other for the target protein, exploiting the intracellular ubiquitin-proteasome system to selectively degrade target proteins.
